Description

  • This antibody is predicted to react with bovine, chicken, amphibian, mouse and rat based on sequence homology
  • Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min
  • Following antigen retrieval, incubate samples with primary antibody for 10 min at room temperature
  • A suggested positive control is tonsil tissue
  • Poly ADP-Ribose Polymerase (PARP) uses nicotinamide adenine dinucleotide (oxidized form) NAD as a substrate to catalyse the transfer of ADP-ribose to a variety of nuclear protein acceptors
  • Proteolysis of PARP to its stable 85kDa fragment is an early marker of programmed cell death (apoptosis) and is mediated by the caspase CPP32 protein
  • Cleavage occurs between Adp216 and Gly217, a site in PARP conserved across species.
