Description

  • Description: The eBioH4B4 monoclonal antibody reacts with human CD107b, also known as lysosomal-associated membrane protein-2 (LAMP-2)
  • CD107b is a highly glycosylated, type I transmembrane protein of approximately 105 kDa
  • It is expressed intracellularly in lysosomal/endosomal membranes in nearly all cells
  • It is also expressed on the surface of degranulating T cells (to a lesser extent than CD107a) and activated platelets as well as some cancer cells
  • In humans, mutations in CD107b results in a lysosomal glycogen storage disorder, known as Danon disease
  • Applications Reported: This eBioH4B4 antibody has been reported for use in flow cytometric analysis
  • Applications Tested: This eBioH4B4 antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product No
  • 88-8824-00) and protocol
  • Please refer to Staining Intracellular Antigens for Flow Cytometry, Protocol A: Two step protocol for intracellular (cytoplasmic) proteins
  • This may be used at 5 μL (0.25 μg) per test
  • A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL
  • Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test
  • eFluor 450 is an alternative to Pacific Blue
  • eFluor 450 emits at 446 nm and is excited with the violet laser line (405 nm)
  • LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b) is a member of a family of membrane glycoproteins
  • LAMP2 provides selectins with carbohydrate ligands, and may play a role in tumor cell metastasis, and have a function in the protection, maintenance, and adhesion of the lysosome
  • Prior to posttranslational modification, LAMP2 is approximately 46 kDa polypeptide
  • Mature, functional LAMP2 is extensively glycosylated with a variety of different N-linked and O-linked oligosaccharides with a total molecular weight of about 100-130 kDa
  • LAMP 2 has a structure comprised of a large amino-terminal intra-lysosomal domain, a hydrophobic trans-membrane domain, and a short carboxyl-terminal cytoplasmic tail
  • LAMP2 is used to monitor a very selective form of autophagy called chaperone-mediated autophagy
  • LAMP 2 has a structure comprised of a large amino-terminal intra-lysosomal domain, a hydrophobic trans-membrane domain, and a short carboxyl-terminal cytoplasmic tail
  • The LAMP protein has 3 variant forms: LAMP-2A, -2B, and 2C
  • LAMP2, together with CD107a/LAMP-1, is a major constituent of lysosomal membrane
  • The LAMP proteins are involved in lysosome biogenesis and are required for fusion of lysosomes with phagosomes, and LAMP2 is an important regulator in successful phagosomal maturation
  • LAMP2 deficiency causes an accumulation of autophagosomes in many tissues leading to cardiomyopathy and myopathy (Danon’s disease).
