Description

  • Description: This monoclonal antibody 47.7G7.1F2 reacts with human CD126, also known as IL-6 receptor alpha
  • It associates with the gp130 chain to form the high-affinity receptor for IL-6
  • Although gp130 is ubiquitously expressed, CD126 is only found on mature T cells, activated B cells, macrophages, hepatocytes, and myeloma cells
  • Upon binding to IL-6, two heterotrimeric complexes of receptor and ligand associate into a hexamer and are then internalized, initializing Jak/STAT signaling pathways
  • IL-6 induces the proliferation and survival of T cells, Th2 differentiation, and acute-phase protein production in the liver
  • A soluble form of CD126 lacking the transmembrane domain is also present and can be generated by cleavage at the cell surface or alternative splicing
  • Soluble CD126 can bind IL-6 and activate cells expressing only gp130 through trans-presentation
  • Applications Reported: This 47.7G7.1F2 antibody has been reported for use in flow cytometric analysis
  • Applications Tested: This 47.7G7.1F2 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells
  • This can be used at 5 μL (0.06 μg) per test
  • A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL
  • Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test
  • Interleukin 6 receptor (IL6R), also known as CD126 (Cluster of Differentiation 126), is a type I cytokine receptor
  • This gene encodes a subunit of the interleukin 6 (IL6) receptor complex
  • Interleukin 6 is a potent pleiotropic cytokine that regulates cell growth and differentiation and plays an important role in the immune response
  • The IL6 receptor is a protein complex consisting of this protein and interleukin 6 signal transducer (IL6ST/GP130/IL6-beta), a receptor subunit also shared by many other cytokines
  • Dysregulated production of IL6 and this receptor are implicated in the pathogenesis of many diseases, such as multiple myeloma, autoimmune diseases and prostate cancer
  • Alternatively spliced transcript variants encoding distinct isoforms have been reported
  • A pseudogene of this gene is found on chromosome 9.
