Description

  • Description: The A2F10 monoclonal antibody reacts with mouse CD135, also known as Flk2/Flt3
  • A member of the tyrosine kinase receptor family, this 135-150 kDa molecule is expressed by primitive progenitor cells in fetal liver and adult bone marrow
  • Two-color staining of adult mouse bone marrow cells with A2F10 and RA3-6B2 (CD45R/B220) or M1/70 (CD11b) reveals Flk-2-positive subpopulations of B cells or myeloid cells, respectively
  • Applications Reported: The A2F10 antibody has been reported for use in flow cytometric analysis
  • Applications Tested: This A2F10 antibody has been tested by flow cytometric analysis of mouse bone marrow cells
  • This can be used at less than or equal to 1 μg per test
  • A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL
  • Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test
  • It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest

  • FLT3 is a tyrosine protein kinase that is a member of the growth factor receptor tyrosine kinase family
  • The receptor is activated by binding of the fms-related tyrosine kinase 3 ligand to the extracellular domain, which induces homodimer formation in the plasma membrane leading to autophosphorylation of the receptor
  • The activated receptor kinase subsequently phosphorylates and activates multiple cytoplasmic effector molecules in pathways involved in apoptosis, proliferation, and differentiation of hematopoietic cells in bone marrow
  • Mutations that result in the constitutive activation of this receptor result in acute myeloid leukemia and acute lymphoblastic leukemia.
