  • Derlins are integral ER membrane proteins that form homo-oligomers and heteromultimeric complexes with membrane-bound machinery, including the E3 ubiquitin ligase Hrd1 and its binding partner SEL1L, involved in ER-associated degradation (ERAD)
  • Before ERAD substrates can be processed by the cytoplasmic ubiquitin-proteasome system, they need to be translocated across the ER membrane through an AAA ATPase p97/VCP-driven process
  • Derlins are inactive members of the rhomboid family of intramembrane proteases that mediate the recruitment of AAA ATPase p97/VCP through interaction via their C-terminal SHP boxes
